The immunological and granulomatous reactions developed during S. mansoni infection are mediated by the activities of both Th1 and Th2 lymphocytes. In this study the kinetic of Th1 and Th2 activities were detected by production of either IFN-g or IL-5, respectively by ELISA and ELISPOT assay, at different times after priming with different schistosomal antigens. Draining lymph node (LN) cells of primed mice were recovered and tested for their ability to proliferate and secrete IFN-g (Th1 activity) and IL-5 (Th2 activity). At day 3 post priming the response was Th1-like, while at day 15 was Th2 -like response. Soluble worm antigen preparation (SWAP) induced marked Th1 activity than Th2 activity while soluble egg antigen (SEA) induced Th2 activity, higher than that of Th1 activity, which was restricted to the first 3 days after priming indicating the important role of SEA in down-regulation of immune response in murine schistosomiasis. |
